A NEW METHOD OF FAT STAINING BY SUDAN ¥²


Abstract
There are many reports dealing with Sudan dye staining, but few method show good results.
Recently Shin in our laboratory revised a new good method and he named it "Sudan III hydrosol method"
This method is highly appreciable for detection of minute fat stricture,
however, for fat rich organ the staining ability by his method weaker than by Sudan III 70% alcohol method. Author intended to overcome this shortening and have done much improve.
After all to compare many Sudan III staining methods, Daddis-Romeis, Kwamura-Yasaki¢¥s, Kim¢¥s, acetone-and Shin¢¥s method were selected and were performed on the slide of fatty liver from intoxicated rabbits with CCl, and on the slide of adrenal cortex from normal rabbits.
From above comparative test, Shin¢¥s method showed best result.
AUTHER,S MODIFIED STAINING SOLUTION IS PREPARED AS FOLLOWS;
1) Sudan III 0.5~1.0gm
¥á-naphthol 0.5~1.0gm
95%-100% alcohol 50cc
Put this solution on the warm water bath for 30 minutes and keep in ice box for 24 hours. Add 95%~90% alcohol to the supernant till the amount become 50cc and add also several droplets of 1 % NaOH solution.
2) Add 50cc Acetone to the precipitation and then the precipitation almost melts away.
Make mixture of same amount Ist solution and II nd solution and add also same amount of 1%gelation solution.
Put on the last mixture on the warm water bath till alcohol and acetone evaporate completely.
THE RESULTS WERE AS FOLLOWS;
1) The finding of slide were exclusively good results that is no precipitation of pigment, no release of fat even long staining duration and distinct staining.
2) It was possible to keep author¢¥s staining solution far more longer than Kim¡¯s method.
3) Among various the above mentioned various methods, Kim¡¯s and Daddis method were satisfiable to get general survey of fat staining.
